Description

A Src-associated protein tyrosine phosphatase (PtpnD1/Ptpn21) is associated with AKAP121 and its splice variant AKAP88 complex in the mitochondrial or cell membrane that helps in focusing and enhancing the cAMP-signaling in the organelle. This signaling complex containing AKAP121, PKA, PTPD1, and src is assembled in vivo. PTPD1 activates src tyrosine kinase and increases the magnitude and duration of epidermal growth factor (EGF) signaling and cause an EGF receptor phosphorylation and downstream activation of ERK 1/2 and Elk1- dependent gene transcription are enhanced by PTPD1. The protein encoded by the ptpn gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an N-terminal domain, similar to cytoskeletal- associated proteins including band 4.1,ezrin, merlin, and radixin. This PTP was shown to specially interact with BMX/ETK, a member of Tec tyrosine kinase family characterized by multimodular structures including PH, SH3, and SH2 domains. The interaction of this PTP with BMX kinase was found to increase the activation of STAT3, but not STAT2 kinase. Studies of the similar gene in mice suggested the possible roles of this PTP in liver regeneration and spermatogenesis (1). The Ptpn21 is a 135 kDa protein (1117 aa) soluble protein with multiple functional domains with N-terminal sequence homology to the ezrinband 4.1-merlin-radixin protein family members. The expression of Ptpn was noted in a variety of tissues; the Ptpn is efficiently phosphorylated by and associated with the src kinase pp60src (2). Recent data indicate that PTPD1 and PTPD2 are expressed in a variety of tissues. In transient coexpression experiments PTPD1 was found to be efficiently phosphorylated by and associated with the src kinase pp60src. PTPD1 is a positive regulator of src signaling and a key component of the EGF transduction pathway. Upon binding of PtpnD1 and or targeting the phosphatase on mitochondria, AKAP121 modulates the amplitude and persistence of src-dependent EGF transduction pathway and established a functional interaction between AKAP and protein tyrosine phosphatase. The Ptpn-selective antibodies were generated against synthetic peptide corresponding to residues 442-460 of the mouse protein tyrosine phosphatase. This sequence is common in rat, and has one mismatch and 2 conservative substitutions in human Ptpn protein. The antibodies to Ptpn are affinity purified over immobilized antigen based affinity chromatography. The purified antibodies are stabilized in antibody stabilization buffer containing preservatives.
